Lung Cancer Diagnosis–Digital PCR And Liquid Biopsy

With the rapid development of precision medicine, many oncogenic driver genes have been identified in non-small cell lung cancer (NSCLC), and molecular typing is a prerequisite for the implementation of targeted therapy in NSCLC. It is clinically important to select accurate, rapid and appropriate assays to comprehensively screen the target population for applicable targeted drugs.
Fusion genes are one of the important types of tumor driver genes that occur in a variety of tumors, and the corresponding targeted drugs have been developed very successfully. The detection of fusion genes has profoundly changed the pattern of tumor typing research and clinical practice, but the difficulty of fusion gene detection has raised higher requirements for detection methods.
Digital PCR liquid biopsy: more sensitive and practical for plasma EGFR mutation detection.
EGFR、ALK、ROS1、MET、HER2、BRAF、RET、KRAS、NTRK……. MET exon 14 skipping mutations are also frequently detected in lung adenocarcinoma and occur in 3-4% of patients. Patients that are positive for MET exon 14 skipping mutations show increased sensitivity to MET inhibitors, including crizotinib and cabozantinib.

Liquid Biopsy of EGFR

The ddPCR assay showed superior sensitivity in the detection of EGFR mutations. This means more patients can benefit from the assay.
ALK/ROS1/MET gene mutation/fusion detection
Two-tube testing:
Assay (A): All types of ALK fusions with unknown partners (expression imbalance).
Assay (RM): 15 common ROS1 fusions, MET exon 14 skipping
